摘要  目的:研究不同浓度龙血素A对瘢痕疙瘩成纤维细胞增殖、凋亡、迁移能力的影响以及对Col-Ⅰ、Col-Ⅲ、α-SMA、Bax、Caspase-3蛋白表达水平的影响。方法:1)不同浓度龙血素A作用于瘢痕疙瘩成纤维细胞,用CellTiter-Blue活力检测试验检测龙血素A对瘢痕疙瘩成纤维细胞增殖的影响。2)流式细胞术检测龙血素A对瘢痕疙瘩成纤维细胞凋亡的影 响。3)划痕实验检测龙血素A对瘢痕疙瘩成纤维细胞迁移的影响。4)Western-Blot测定龙血素A对瘢痕疙瘩成纤维细胞Col-Ⅰ、Col-Ⅲ、α-SMA、Bax、Caspase-3蛋白水平的影响。结果:龙血素A可抑制瘢痕疙瘩成纤维细胞增殖、迁移并诱导其凋亡,同时减少COL-Ⅰ、COL-Ⅲ、α-SMA蛋白表达,并促进Bax、Caspase-3蛋白表达,并且均呈现剂量依赖性。结论:龙血素A可以调控瘢痕疙瘩成纤维细胞增殖与凋亡的平衡、减少细胞外基质沉积,从而可能对KD发挥一定的治疗作用。

Study on the Effects of Loureirin A on Keloid Fibroblast Growth Function

Abstract  Objective:To study the effects of different concentrations of Loureirin A on the proliferation, apoptosis and migration of keloid fibroblasts and the protein expression levels of Col-, Col-Col-Ⅰ,α-SMA, Bax, Caspase-3. Methods:1)Different concentrations of L-Auxin A act on keloid fibroblasts. CellTiter-Blue vitality test was used to test the effects of L-Auxin a on keloid fibroblast proliferation. 2)Flow cytometry was used to detect the effects of L-Auxin A on keloid fibroblasts apoptosis. 3)Scratch test was used to detect the effects of borneol A on keloid fibroblast migration. 4)Proteins expression levels of Col-Ⅰ, Col-Ⅲ, α-SMA, Bax Caspase-3 were detected by Western-Blot. Results:Loureirin A can inhibit the proliferation and migration of keloid fibroblasts, an induce their apoptosis, reduce the protein expression of Col-Ⅰ, Col-Ⅲ α-SMA and promote the protein expression of Bax and caspase-3, and there is a dosage dependent relationship. Conclusion:Loureirin A can regulate the balance of keloid fibroblast proliferation and apoptosis, reduce the deposition of extracellular matrix, so it may play a certain therapeutic effects on keloid.
